Pharmacokinetics
When administered orally, quetiapine is well absorbed and actively metabolized. Eating does not significantly affect the bioavailability of quetiapine. Quetiapine is approximately 83% bound to plasma proteins. Geroquel maximum molar concentration of the active metabolite of N-desalkyl-quetiapine in equilibrium is 35% of the noted concentrations for quetiapine. The half-lives of quetiapine and N-desalkyl-quetiapine are about 7 and 12 hours, respectively.
The pharmacokinetics of quetiapine and N-desalkyl-quetiapine in the approved dose range is linear. In men and women, the kinetics of quetiapine does not differ.
The average clearance of quetiapine in elderly patients is approximately 30–50% lower than in adult patients aged 18 to 65 years.
In subjects with severe impaired Geroquel function (creatinine clearance less than 30 ml / min / 1.73 m2), the average plasma clearance of quetiapine was reduced by about 25%, however, some indicators of clearance corresponded to the range for healthy subjects.
