Overview
It is available in several dosage forms: buprenorphine in ampoules with a solution for intravenous and intramuscular injection (0.03% in 1 ml), sublingual tablets of buprenorphine (0.2 g of buprenorphine hydrochloride in a package of 10 to 100 pcs) and in blisters.
Mechanism of action
The speed of the drug is different from the form of release. With intravenous administration, the effect is observed less than in a minute and reaches its maximum concentration in 20 minutes. With intramuscular - it is felt after 15 minutes. With sublingual - after 30 minutes.
The analgesic effect lasts from six to eight hours. The maximum dosage is 1.2-1.5 mg, but respiratory failure begins.
You can not grind the Buprelieve in the form of tablets and breed it for injection. This not only does not improve the result, but also leads to death.
Thirty times stronger than morphine buprenorphine
The analgesic effect of buprenorphine is almost thirty times higher than morphine, so 0.3 mg of this drug is 10 mg of morphine.
After administration, a strong analgesic effect and mild euphoria occur. Moreover, it is less toxic and less often highly addictive.
The mechanism of action of buprenorphine, like other agonists, is aimed at interacting with mu- (μ-) and kappa- (κ-) opioid receptors, which is the reason for the decrease in pain and satisfaction.
At the same time, the connection of the drug with μ receptors is strong, because of which the suppression of all other agonists.
Own activity with μ receptors is weak, but with κ receptors not found. Therefore, buprenorphine can be attributed to Buprelieve group of partial antagonists that contribute to treatment for opioid addiction.
Buprenorphine abuse
Semi-synthetic opioid causes negative consequences if the patient has a number of diseases, for example:
- respiratory failure (asthma, chronic obstructive pulmonary disease);
- hepatitis B, C;
- kidney and thyroid problems;
- deviations in the work of the stomach;
- pathology of the gallbladder;
- violations in the activity of the adrenal glands;
- mental illness.
